NAD+ Therapy: What the Science Actually Says

NAD+ has become one of the most talked-about compounds in the longevity and wellness space. The enthusiasm around it is occasionally overblown and the marketing often strays from what the science actually supports. What NAD+ Is

Nicotinamide adenine dinucleotide (NAD+) is a coenzyme found in every living cell. It plays a central role in several biological processes:

Cellular energy production. NAD+ is essential to the mitochondrial reactions that convert nutrients into adenosine triphosphate (ATP), the primary energy currency of the cell. Without adequate NAD+, cellular energy production becomes less efficient.

DNA repair. NAD+ is a required substrate for PARP enzymes, which are involved in detecting and repairing DNA strand breaks. This process is ongoing in every cell and becomes more relevant as the pace of DNA damage outpaces repair capacity.

Sirtuin activation. Sirtuins are a family of proteins involved in cellular stress responses, metabolic regulation, and aging-related pathways. They require NAD+ to function. Research into sirtuins has driven much of the commercial interest in NAD+.

Circadian rhythm regulation. NAD+ levels fluctuate across the day and are tied to circadian biology, which affects sleep quality, metabolic function, and cellular repair cycles.

What Happens to NAD+ Levels Over Time

NAD+ levels decline with age. Research has consistently documented this decline beginning in early adulthood and continuing progressively. Some studies have shown reductions of 40 to 60 percent between young adulthood and middle age.

This decline is associated with reduced mitochondrial efficiency, impaired DNA repair, and changes in metabolic function. Whether restoring NAD+ levels through supplementation reverses these changes in humans is still an active area of research, but early findings in several areas are encouraging.

Why IV Delivery

Oral NAD+ precursors such as NMN (nicotinamide mononucleotide) and NR (nicotinamide riboside) are converted to NAD+ through enzymatic pathways in the body. They are effective at raising NAD+ levels and are widely studied.

IV NAD+ delivers the compound directly into the bloodstream without conversion requirements. This produces a faster and more significant elevation in circulating NAD+ levels, which is the mechanism by which patients sometimes notice pronounced effects within hours of a session. IV delivery also bypasses the variability in oral absorption that can limit the predictability of supplement-based protocols.
